In 1958, "Symphony" was composed by the commission of Japan Philharmonic Orchestra. In 1960, "Cello Concerto" was composed by the commission of NHK Symphony Orchestra. In 1965, "Counterpoint" was published. In 1968, "Piano Concerto" won the 16th Otaka Prize and Ministry of Education Festival Encouraging Award. In the same year, he became an assistant professor at Tokyo University of the Arts. In 1974, he became a professor at Tokyo University of the Arts. In addition, he composed some school songs, and the last one is a song of Nabari Kikyo-ga-oka High School.
On April 9, 1976, during composing "Violin Concerto", he died suddenly at the age of 46 due to heart failure. The cemetery is located at the Jodo Buddhist temple in Higashi Kanagawa, Yokohama City, Kanagawa Prefecture.

His death was so premature that he was deification from early, and his personal complete works were planned and published shortly after his death.
Dan Ikuma, who is Japanese composer, described the scene when he met Akio (when Yashiro was an elementary school student, and Dan was a junior high school student), the exchanges that followed, and when he contacted the news of Yashiro's death at "Cloud Procession" (pp. 58-63) in his book "Kasanete Pipe-no-kemuri or the Smoke of Pipes again" (Asahi Shimbun-sha, 1980). In this book, he faithfully wrote that he happened to know Yashiro's death in Beijing on the day after he died, his death was because of fatigue of professor's works, and Toru Takemitsu sharply told Teizo Matsumura, "Quit the university, Mr. Matsumura. We should just compose." These are a record that tells the shock of Japanese composers when Yashiro died. Takemitsu was sometimes asked by Yashiro to become an instructor at the University of the Arts, but he never taught at a music school.

1958年、日本フィルハーモニー交響楽団の委嘱により「交響曲」を作曲。1960年、NHK交響楽団の委嘱により「チェロ協奏曲」を作曲。1965年、「対位法」を出版。1968年、「ピアノ協奏曲」が第16回尾高賞・文部省芸術祭奨励賞を受賞。同年、東京藝術大学助教授に就任。1974年、東京藝術大学教授となる。そのほかにも、幾つかの高校の校歌も作曲しており、最後の校歌としては、名張桔梗丘高校の校歌を作っている。
ヴァイオリン協奏曲を作曲中の1976年4月9日、心不全により46歳で急逝。墓所は、神奈川県横浜市東神奈川にある浄土宗成仏寺の墓地にある。

あまりにも早すぎた死により、早くから神格化され、没後すぐに個人全集が企画・出版されるなど、異例の待遇で評価された。
團伊玖磨はその著書『重ねて・パイプのけむり』（朝日新聞社・昭和55年（1980年）1月30日第1刷発行）の「雲の行列」（同書58～63ページ）で、矢代秋雄との出会い（矢代が小学生、團が中学生の時）や、その後の交流、そして矢代秋雄の死の報に接した際の光景を綴っている。團は矢代の死を死の翌日、第1回日中文化交流協会音楽家代表団の一員（団長）として滞在中の北京で偶然知り、矢代の死因が作曲を続けながら芸大主任教授を務めていたことによる疲労が原因であったこと、また、同じく代表団で同席していた武満徹が松村禎三に対し、「松村君、芸大なぞは辞めなさい。作曲だけをしよう」と鋭く言ったこと等を生々しく記している。矢代が死去した当時の日本の作曲家の衝撃を語る記録である。武満徹は矢代から芸大講師に就任を打診されたこともあるが、終生、音楽学校での教鞭を執ることがなかった。
